Lactobacillus helveticus R0052 and Bifidobacterium longum R0175 Supplementation: An Exploratory, Randomized, Placebo-Controlled Trial of Endocannabinoid and Inflammatory Responses in Female Dancers
Verified on PubMed Oxford CEBM Level 1B: Randomized Controlled Trial
A 2025 randomized controlled trial in Microorganisms tested endocannabinoid system in inflammation. It made no clear difference. Neither the bacteria capsule nor the dummy capsule produced changes in the chemicals or stress measures that the researchers could confidently tell apart from chance. This study does not show that this probiotic affects the body's natural calming.
In plain language
Study overview
This study looked at young women who train as professional dancers. Some took a daily probiotic supplement, which is a capsule of helpful bacteria, and others took a look-alike sugar-and-starch capsule for three months. The researchers checked the body's natural calming chemicals and signs of inflammation before and after.
Findings
It made no clear difference. Neither the bacteria capsule nor the dummy capsule produced changes in the chemicals or stress measures that the researchers could confidently tell apart from chance.

The deeper readScientific analysis
The anandamide (AEA) and lipopolysaccharide (LPS) interaction is gaining attention, but evidence on the influence of probiotics on endocannabinoid system (ECS) biomarkers remains limited. This study (NCT05567653) investigated the effects of 12-week supplementation with Lactobacillus helveticus R0052 and Bifidobacterium longum R0175 on AEA (main outcome) and inflammatory biomarkers in female dancers. Fifteen participants (5 probiotic, 10 placebo) were included in the final analysis. Serum levels of AEA, LPS, and cytokines (TNF-α, IL-1β, IL-10) were measured using ELISA, and psychological stress responses were evaluated using the Mini-COPE questionnaire. At baseline, a correlation between AEA and LPS was observed (Spearman's r = 0.9677, p < 0.05). After 12 weeks, no statistically significant differences in AEA, LPS, cytokine levels, or stress-coping strategies were observed between the probiotic and placebo groups (LPS–probiotic: +3.48 EU/L, p = 0.9361; placebo: +56.98 EU/L, p = 0.0694; AEA–probiotic: −1.11 ng/mL, p = 0.9538; placebo: +14.08 ng/mL, p = 0.4749). The direction of change may indicate a trend toward increased inflammation in the absence of probiotics. These results should be viewed as hypothesis generating and warrant confirmation in larger trials.

Limitations the authors noted
Only 15 of 51 targeted participants analyzed; study underpowered (power analysis indicated >100 needed). All participants female; results not generalizable to males. Post-randomization exclusion of two participants without intention-to-treat analysis. ELISA used for AEA quantification, less specific than LC-MS/MS. No sedentary control group. Gut microbiota composition not analyzed. Dietary intake assessed only at baseline. Industry-affiliated authors (probiotic company shareholder and paid consultant). High attrition (dropout ~41%). Assay CV (<10–12%) may mask subtle effects.
In practice
Why this matters
This study does not show that this probiotic affects the body's natural calming system or inflammation, because the group was very small and many women dropped out. It does suggest that a larger, longer study would be needed before anyone can draw a conclusion about these bacteria in active people. Readers should not take it as a reason to start or avoid any supplement, and results from one small group of female dancers may not apply to others.
